ABSTRACT. Banana (Musa spp.) is one of the main fruits consumed worldwide. However, finger drop, is a physiological disorder that causes many postharvest problems, which eventually reduces market value and consumer acceptance. Therefore, the objective of the study was to evaluate the anatomical changes that occur in the pedicel rupture area (drop zone) of bananas diploids (BB França) and tetraploid (BRS Pioneira) in three ripening stages. The levels of gene expression involved in the natural ripening process and in the development of finger drop, was also investigated. The accumulation of their mRNAs and those of expansin (EXP1), pectate lyase (PEL1) and xyloglucan endotransglucosylase/hydrolase protein (XTH4) genes already isolated from bananas were measured by quantitative polymerase chain reaction in three ripening stages. BB França presented a higher resistance to finger drop due to the presence of some specific morphoanatomical characteristics, such as larger parenchymal cells and greater deposition of lignin. In contrast, there was degeneration of the pedicel parenchymal tissue of the BRS Pioneira genotype, forming large empty spaces during the ripening of the fruits, mainly in stage 6, which contributed to the finger drop. The diploid BB França is a strong candidate for use in banana breeding programs aimed at fruit drop resistance. This will certainly improve the quality of banana varieties. Moreover, PEL1 proved to be an excellent candidate gene for functional studies of finger drop in bananas.

RESUMO A toxicidade do alumínio (Al) em plantas demonstra que a avaliação de genótipos é importante para o reconhecimento de marcadores de tolerância. Objetivou-se avaliar os efeitos do estresse por Al no teor relativo de água, danos de membrana e alterações anatômicas, em variedades de girassol tolerante e sensível ao Al. Plantas de girassol [Catissol (tolerante) e IAC-Uruguai (sensível)] foram cultivadas em solução nutritiva (controle) ou solução nutritiva com 0,15 mM de AlCl3 (tratamento de estresse), em casa-de-vegetação. O delineamento experimental foi o inteiramente casualizado, em esquema fatorial composto por quatro épocas de colheita x duas variedades de girassol x dois níveis de Al, com quatro repetições. Os resultados demonstraram efeitos deletérios do Al na porcentagem de integridade absoluta e no teor relativo de água apenas na variedade IAC-Uruguai. Estes resultados nas folhas estressadas da variedade sensível podem ter advindo dos danos na estrutura do xilema. Além disso, o aumento da espessura da lâmina foliar e nas camadas de parênquima, bem como a lignificação de tecidos da raiz, são características importantes de plantas IAC-Uruguai, podendo ser utilizadas como marcador anatômico da sensibilidade do girassol ao Al.

ABSTRACT Aluminum (Al) toxicity in plants evidences the importance of genotype evaluation to the identification of tolerance markers. This study aimed at evaluating the effects of aluminum stress on the relative water content, membrane damages and anatomical changes, in Al-tolerant and Al-sensitive sunflower cultivars. Sunflower plants [Catissol (Al-tolerant) and IAC-Uruguai (Al-sensitive)] were grown in nutrient solution (control) or nutrient solution containing 0.15 mM of AlCl3 (Al-stress treatment), in a greenhouse. The experimental design was completely randomized, in a factorial arrangement consisting of four harvest times x two sunflower cultivars x two Al levels, with four replications. The results showed that Al negatively affected the absolute integrity percentage and relative water content only for the IAC-Uruguay cultivar. These results in the stressed leaves of the Al-sensitive cultivar may be due to damage in the xylem structure. In addition, the increase in leaf blade thickness and parenchyma layers, as well as lignification of root tissues, are important traits of IAC-Uruguay plants and may be used as anatomical markers of Al sensitivity in sunflower.

The structure and histochemistry of colleters found on the vegetative and floral apices of Odontadenia lutea are described. Colleters occur on vegetative apices starting at the fourth node, with 68 to 80 colleters being found at each node. Each leaf primordium has only one colleter of axillary origin, 3-5 intra-petiolar, and 12-16 inter-petiolar (intra-stipular). There are four types of colleters: standard, bipartite standard, sessile, and bipartite sessile. Colleters on the reproductive apices alternate with the sepals and are sessile, reduced sessile, tripartite laminar sessile, or asymmetrical. All of the colleters have a central nucleus of parenchymatous cells covered by a palisade uniseriate secretory epidermis and a thin cuticle. Secretory idioblasts were observed in the parenchymatous axis. Vascularization was observed only in standard axillary and laminar colleters. Crystals were observed in the parenchyma of the axillary colleter. Histochemical tests demonstrated that there was no rupturing or distension of the cuticle during the secretion process. Mucilage was identified using the PAS reaction as well as by Mayer's reagent and Ruthenium red staining. The calycine colleters had two distinct secretory phases, the first synthesizing mucilage and the second producing phenolic compounds.

Resumo Este trabalho teve como objetivo descrever a ontogenia do pericarpo de Temnadenia violacea (Vell.) Miers. O ovário é glabro, súpero, bicarpelar e de placentação sutural. Cada carpelo apresenta um único lóculo onde se dispõem vários óvulos inseridos em um tecido placentário bem desenvolvido. A parede do ovário é composta de epiderme externa, mesofilo ovariano e epiderme interna. O fruto é um folicário constituído de dois frutículos geminados, cilíndricos, alongados e lenhosos. Nesse estudo foram consideradas duas fases distintas de desenvolvimento: fruto jovem e fruto maduro. No fruto jovem, o epicarpo é unisseriado e o mesocarpo pode ser dividido em duas regiões distintas. O endocarpo é formado por células alongadas em seção transversal. No fruto maduro o epicarpo é recoberto por uma cutícula espessa e papilosa, no mesocarpo a última camada de células próximas ao endocarpo torna-se esclerificada e juntamente com as células do endocarpo formam o endocarpo funcional. No mesocarpo do fruto maduro foram observados laticíferos não articulados e ramificados, fibras lignificadas e fibras não lignificadas.

Abstract This work describes the ontogeny of the pericarp of Temnadenia violacea (Vell.) Miers. The ovary is glabrous, superior, with bicarpelar sutural placentation. Each carpel has a single locule with several eggs attached to a well-developed placenta. The ovary wall is composed of outer epidermis, mesophyll and inner epidermis. The fruit is a follicarium composed of two geminate aggregate fruits that are cylindrical, elongated, and woody. In this study two distinct phases of development were considered: young fruit and ripe fruit. The epicarp of the young fruit is uniseriate and the mesocarp can be divided into two distinct regions. The endocarp is formed by cells that are elongated in transverse section. The epicarp of the mature fruit is covered by a thick cuticle having papillae, while the layer of mesocarp cells nearest the endocarp become sclerotized - and together with the cells of the endocarp form the functional endocarp. Non-articulated and ramified laticifers, and lignified and non-lignified fibers were observed in the mesocarp of the mature fruit.

(Coléteres foliares e calicinais de Temnadenia violacea (Apocynaceae, Apocynoideae): estrutura e distribuição). Este trabalho descreve a origem, estrutura e posição dos coléteres dos ápices vegetativos e florais de Temnadenia violacea (Vell.) Miers e comprova a presença de mucilagem na secreção produzida por estas estruturas. O número de coléteres foliares varia de 9 a 11 por primórdio e de 18 a 22 por nó; apenas um tem origem axilar, sendo os demais de origem marginal. Quanto à posição, cinco coléteres são peciolares e os demais interpeciolares. Os coléteres foliares são dos tipos standard e séssil, sendo constituídos por uma porção alongada, formada por um núcleo central de células parenquimáticas, revestido por epiderme secretora em paliçada uniestratificada e cutícula delgada. Tricomas tectores e tecido vascular ocorrem apenas nos coléteres marginais distais. No ápice floral, os coléteres calicinais têm origem na base do cálice, sendo três coléteres opostos a cada uma das lacínias. Todos os coléteres calicinais possuem um núcleo central de células parenquimáticas, epiderme secretora em paliçada uniestratificada, cutícula delgada e são sésseis; o eixo parenquimático destes coléteres é destituído de vascularização e os laticíferos observados são de pequeno calibre. A mucilagem foi detectada tanto na secreção dos coléteres foliares quanto dos calicinais.

(Foliar and calycine colleters of Temnadenia violacea (Apocynaceae, Apocynoideae): structure and distribution). The present work describes the origin, structure and position of the colleters on vegetative and floral apices of Temnadenia violacea (Vell.) Miers and confirms the presence of mucilage in the secretion produced by those structures. The number of foliar colleters ranges from 9 to 11 per primordium and from 18 to 22 per node; only one has an axilar origin, while the others originate along the margins. Concerning position, five of them are petiolar and the others are interpetiolar. There are two types of foliar colleters: standard and sessile, and they consist of a main body composed of a central core of elongated parenchyma cells surrounded by a secretory palisade epidermis and a thin cuticle. Tector trichomes and vascular tissue occur only on the distal marginal colleters. On the floral apices, the calycine colleters are formed at the base of the calyx, three of them opposite the sepals. All of the calycine colleters have a central core of elongated parenchyma cells, a secretory palisade epidermis, a thin cuticle, and are sessile. The calycine colleters are not vascularized and the laticifers are narrow. Mucilage was detected in the secretion of both foliar and calycine colleters.

As técnicas usuais empregadas no processamento de materiais herborizados são limitantes porque não permitem reproduzir um laminário permanente e porque o registro dos resultados é temporário. Em estudos de anatomia aplicada à taxonomia, muitas vezes, é necessário utilizar exsicatas. Este trabalho teve como objetivo testar a utilização de metacrilato (Historesin, Leica) na preparação e inclusão de material herborizado, visando a obtenção de laminário permanente. Foram utilizadas folhas de plantas herborizadas do gênero Senecio Tourn. ex L. (Asteraceae), previamente reidratadas em H2O e armazenadas em etanol 70%. As amostras foram desidratadas em série etanólica e submetidas à (1v:1v) mistura de AE 95% e resina pura. Durante a infiltração, com resina pura, o material foi submetido a vácuo durante 72 horas. Os blocos foram cortados em micrótomo de avanço automático, com navalhas de aço descartáveis, e os cortes foram corados com azul-de-toluidina (pH 4,0). As lâminas foram montadas em Permount. O laminário permanente obtido apresentou qualidade superior à dos produzidos pelas técnicas usuais, pois foi possível obter cortes seriados, com espessura determinada. Essa técnica possibilitou otimizar a obtenção de cortes, a partir de pequenos fragmentos de exsicatas.

The usual herbalized material processing techniques are limited due to the difficulties to produce permanent slide collections, and to register permanent results. In studies of taxonomy-applied anatomy, it is often necessary to use herbarium specimens. The objective of this study was to test methacrylate (Historesin, Leica) in the preparation and inclusion of herbalized materials to obtain a permanent slide collection. Herbalized leaves from the genus Senecio Tourn. ex L. (Asteraceae) were used, after being submitted to re-dehydration in ethanol 70%. The samples were dehydrated in an ethanol series and submitted to a mixture of AE 95% plus pure resin (1v:1v). The embbeding process in pure resin was submitted to a vaccum during 72 hours. The blocks were cut in an automatic progress microtome, using disposable steel razors and the cuts stained with Toluidine Blue pH 4.0. The slides were mounted in Permount. The permanent slide collection obtained showed high quality, compared with those obtained by the usual techniques, because the cuts were seriate; cut thickness could be determined allowing the use of few samples for the analysis of the material.
